高效和超高效液相色谱法测定鼻舒适片中欧前胡素含量的比较

【摘要】目的运用高效液相色谱法和超高效液相色谱法对鼻舒适片中欧前胡素的含量进行测定，并对测定方法和结果进行比较。
方法高效液相色谱法中，色谱柱采用迪马C18柱（250mm×4.6mm，5μm），流动相为甲醇-水（65：35），流速为1.0 ml/min，检查波长为300nm，进样量为10μl。
超高效液相色谱法中，色谱柱采用Shim-pack XR-ODSIIIC18（70mm×2mm，1.6μm），流动相为甲醇-水（58：42），流速为0.3 ml/min，进样量为3μl。
结果高效液相色谱法主峰出峰时间为45min，3批样品含量分别为0.086,0.085,0.082mg/片。
超高效液相色谱法主峰出峰时间为3.5min，3批样品含量分别为0.087,0.086,0.082mg/片。
结论两种方法均可用于测定鼻舒适片中欧前胡素的含量，与高效液相色谱法相比，超高效液相色谱法加快了分离速度，提高了分离效率，减少了有机溶剂的使用。

【Abstract】 Objective：To determine the content of Imperatorin in Bishushi Tablets by the HPLC and UPLC methods and to compare the detecting methode and their detected results.Methods：In the HPLC method,the Diamonsil C18 column(250 mm×4.6 mm,5μm)was used with the mobile phase of methanol-water(65:35)at a flow rate of 1.0ml/min,the injection volume was 10μl,and the detection wavelength was set at 300nm.. In the UPLC method, Shim-pack XR-ODSIIIC18 column(70 mm×2mm,1.6μm)was used with the mobile phase of methanol-water(58:42)at a flow rate of 0.3ml/min ,and the injection volume was 3μl.Results：The appearance time of the main peak in HPLC was 45min,and the content of Imperatorin in there batches of samples were 0.084,0.082,0.081 mg /Tablet,respectively. 鼻舒适片是由苍耳子、野菊花、鹅不食草、白芷、防风、墨旱莲、白芍等十一味药组成的复方制剂，收载于《卫生部药品标准•中药成方制剂》[1]第七册，具有清热消炎、通窍的功效，用于治疗慢性鼻炎引起的喷嚏、流涕、鼻塞、头疼，过敏性鼻炎，慢性鼻窦炎。
笔者分别采用超高效液相色谱法和高效液相色谱法对鼻舒适片中欧前胡素进行含量测定，并对测定结果进行比较，旨在为鼻舒适片质量控制提供新方法。
1 仪器与试药
岛津LC-30A超高效液相色谱仪，岛津LC-20A高效液相色谱仪；欧前胡素对照品（中国药品生物制品检定所，批号为110826-201013，供含量测定用，含量以99.6%计）；甲醇为色谱纯，水为超纯水，其他试剂均为分析纯；鼻舒适片（广东省博罗先锋药业集团有限公司，批号分别为000045、000057、000069）。
2 方法与结果
2.1 HPLC法[2]
2.1.1 色谱条件
色谱柱：迪马C18柱（250mm×4.6mm，5μm）；流动相为甲醇-水（65：35）；流速为1.0 ml/min；进样量：10μl；检查波长：300nm 。
2.1.2 溶液的制备
精密称取欧前胡素对照品适量，加甲醇制成每1 ml含约20μg的溶液，作为对照品溶液；取本品10片，除去包衣，研细，混匀，称取约0.5g，精密称定，置50 ml具塞锥形瓶中，精密加入甲醇10 ml，密塞，称定重量，超声处理（250W，频率33kHz）1小时，放冷，再称定重量，用甲醇补足减失的重量，摇匀，滤过，取续滤液，作为供试品溶液；取缺白芷的阴性样品0.5g，同供试品溶液制备方法制成阴性对照品溶液。
2.1.3 方法学考察
专属性试验：精密吸取对照品溶液、供试品溶液和阴性对照品溶液各10μl，依法进行测定，结果阴性对照品溶液无干扰（见图1）。
