当代化工研究丄 O I I Chenmical I ntermediate生物制药■与研究2019•01乳腺癌致病基因BRCA1的生物信息学分析*辛艾玲(成都七中实验学校四川610000)摘要:BRCA1作为乳腺癌关键蛋白参与了乳腺癌和卵巢癌的发生,本文对BRCA1基本特性进行了初步的分析,通过分析其整体的亲疏水 性,氨基酸组成,表明BRCA1倾向是核蛋白,整体呈亲水性•使用PS0RT2分析工具对BRCA1亚细胞定位进行领测,表明BRCA1蛋白主要分 布在细胞核中.利用MEGA软件进行BRCA1系统发育分析,结果表明人BRCA1与猿的BRCA1亲缘关系高度相近•最后,采用同源建模(比较建 模)方法构建了BRCA1蛋白的三级结构,从建模质量评估显示预测结果准确。

This p aper makes a preliminary analysis o f t he basic characteristics ofBRCAl. By analyzing its overall hydrophilicity and h ydrophobicity and a mino acid c omposition, it shows that BRCA1 tends to be nucleoprotein and o verall shows hydrophilic. By using P SORT2 analysis tool to cany out the p rediction on the location o fB RCA1 subcells, it indicats that BRCA 1 protein is mainly distributed in the nucleus. By making phylogenetic analysis o f B RCA1 using MEGA software, it s howed t hat the g enetic relationship between human BRCA1 and a pe BRCA1 was highly s imilar. Finally, the three-level s tructure ofBRCAl protein was constructed by the method o f h omologous modeling (comparative modeling), and the p rediction results were accurate f rom the modeling quality evaluation.K ey w ords i BRCA1 protein;Three-level structure p rediction;subcellular localization p rediction;phylogenetic analysis1背景近些年,我国女性乳腺癌的发病几率逐渐升高,特别 是在一线城市的发病率很高。

目前,乳腺癌已位居女性恶性 肿瘤首位,在家族中会发生家族聚集现象,且发病年龄逐渐 年轻化,极大地威胁着我国妇女的身体健康,我们需对此引 起重视。

乳腺癌易感基因(BRCA1)是乳腺癌的危险因素之一,是目前为止发现的与乳腺癌相关的最重要的抑癌基因。

它在遗传性乳腺癌患者中有较高的突变率,已成为评估女性 患乳腺癌风险和指导方案选择的重要分子标志物。

其生物 功能广泛,对细胞周期,基因转录,D N A修复等功能均有影 响。

BRCA1蛋白是一种由BRCA1基因产生的由1863个氨基酸组 成的多功能区核蛋白,它与其它肿瘤抑制因子和信号传感器 等形成复合物,在D N A修复损伤,转录调控,细胞周期的调 节和泛素化方面起着重要作用。

本论文就BRCA1蛋白的氨基 酸组成、疏水性、亚细胞定位、系统发育分析和它的蛋白质结构进行了分析,我们希望这些结果对于更进一步地了解 BRCA1与乳腺癌的关系起到一定作用。
