8544 Abstract BACKGR0UND:Human adipose tissue-derived stromaI cells(ADSCs)possess exert proliferation and multi—directionaI ability.As a novel stem cell,it has been widely utilized in tissue engineering and plays an important role in biological and potential therapeutic applications. OBJECTIVE:To review the research progress,applications and existing problems of human ADSCs in tissue engineering and celt therapy by retrieving relevant publications. METH0DS:PubMed and CNKI dafabases were undertaken to identify the relevant articles published from January 1 960 to January 2009 with the key words of“human adipose tissue—derived mesenchymal cells.isolation,diferentiation.immune phenotype,application”both in English and Chinese.The articles relate to biological characteristics and applications of ADSCs were selected.In the same field,the documents published earlier or in the authoritative journals were preferred,and the repetitive studies were excluded. RESULTS A ND CONCLUSION:A total of 81 documents were searched by computer.and 57 references were incIuded}n the finaI analysis.The results demonstrated that,human ADSCs share many characteristics.including the high proliferative potential and exhibiting the exert ability to undergo multilineage diferentiation under appropriate conditions.Human ADSCs can not only utilize for tissue repairing,but also for cell immune modulation and gene therapy.However.there are still problems in its application With the development of research on human ADSCs.their biological characteristics wIll be revealed.and their application in lissue repairing,cell therapy,transplantation,as well as gene therapy must be hold a great promise INTRODUCTION Preliminarily marrow mesenchymaI stem cells fMSCs1 studies have focused on the bone marrow mesenchymal stem cells(BMSCs).BMSCs can be isolated fr0m bone marrow and expanded in vitro without any apparent modification in phenotype or fOSS of biological function,arid have ability of multi.potent diferentiationll-JJ.These make the BMSCs as the preferred candidate stem cells for mesodermal defect repair and stem celI therapy basement.However the problems of BMSCs applied in clinic are presented.including pain of bone marrow aspiration。morbidity and Iow cell number collected, these disadvantages have forced the many researchers to exploit other ideaI source of MSCs. Human adipose tissue—derived stromaI cells(ADSCs) are a rich source of pluripotent mesenchymaI cells and can be easily obtained by Iiposuction or subcutaneous surgical operation fr0m adipose tissue with minimally invasionIq j.The experimental research indicated that ADSCs possesses high proliferative potential and exhibits abihty to undergo multilineage differentiation.They have the potential to induced to di仟erentiate into not only adipocytes. cartilage,but also bone,skeletaI muscle,neurons and editheliaI cells under appropriate culturing conditions with specific hormones or growth factors[’0— .Due to the abundant quantities.easy collection and isolation operation.ADCSs are becoming a ̄ractive alternative MSCs for treatments Of human disease.such as inherited.traumatic or degenerative bone ioint and soft tissues defect repairing.The aim of this review was to:describe the preparation and isolation procedures of ADSCs; summarize the molecular and immunophenotype features;describe the differentiation of ADSCs;as welI as discuss the future role of ADSCs in tissue engineering repair and stem cell therapy. MATERIALS AND METHODS Data resources PubMed(http://www.ncbi.nlm.nih.gov/pubmed/)and CNKI(http://www.cnki.net/)databases were seamhed by the first author to identify the relevant articles published frOm January 1 960 to January 2009 with the key words of“human adipose tissue-derived mesenchVmal cells,isolation. diferentiation,immune phenotype,application”both in English and Chinese. Selective criteria Inclusive criteria:studies related to the culture method.diferentiation ability.immune characteristics ofADSCs in tissue engineering and cell therapy.In the same field,the documents published earlier or in the authoritative iournals were preferred.Exclusive criteria:obsolete studies,repetitive studies,Meta analysis or unrelated articles. Quality assessment Totally 81 articles were retrieved,including 79 English articles and 2 Chinese articles.According to inclusive and exclusive criteria,57 repetitive researches were excluded.The inclusive papem referred to the method for isolation and culture of ADSCs.the diferentiation ofADSCs.molecular expression and immunophenotype of ADSCs.as weI as application prospects of ADSCs in tissue engineering and stem cell therapy.
