丹瓜方对高糖高脂喂养大鼠Nampt、InsR、PKB的影响摘要:随着现代生活方式的改变,高糖高脂饮食成为人类健康的重大问题之一。

本研究旨在探讨丹瓜方对高糖高脂饮食喂养大鼠Nampt、InsR、PKB的影响。

实验采用53只雄性SD大鼠,随机分为对照组、模型组和丹瓜方组。

模型组和丹瓜方组饲喂高糖高脂饲料4周。

丹瓜方组在饲料基础上加入丹瓜方水煎液,每天灌胃。

实验结束后,采用荧光定量PCR、Western blot和组织病理学方法测定鼠肝脏Nampt、InsR、PKB mRNA和蛋白表达情况,同时记录血糖、血脂变化。

结果发现,与模型组相比,丹瓜方组的血糖、血脂水平明显下降,肝脏中Nampt、InsR、PKB mRNA和蛋白表达水平也有所下调,组织病理学显示肝脏脂肪变性减轻。

因此,丹瓜方在高糖高脂饮食喂养大鼠中可能通过调节Nampt、InsR、PKB表达来缓解高糖高脂饮食带来的代谢异常。

This study aimed to investigate the effect of Dan Guafang on Nampt, InsR and PKB in rats fed with high sugar and high fat diet. Fifty-three male SD ratswere randomly divided into control group, model group and Dan Guafang group. The model group and Dan Guafang group were fed with high sugar and high fat diet for 4 weeks. The Dan Guafang group was administered with Dan Guafang water extract daily by gavage. After the experiment, fluorescence quantitative PCR, Westernblot and tissue pathology were used to measure the mRNA and protein expression of Nampt, InsR and PKB in rat liver, and the changes of blood sugar and blood lipid were recorded. The results showed that compared with the model group, the blood sugar and blood lipid levels in the Dan Guafang group decreased significantly, and the expression levels of Nampt, InsR, and PKB mRNA and protein in the liver were also down-regulated. Tissue pathology showed that liver steatosis was reduced. Therefore, Dan Guafang may alleviate metabolic abnormalities caused by high sugar and high fat diet by regulating the expression of Nampt, InsR and PKB.Keywords: Dan Guafang; high sugar and high fat diet; Nampt; InsR; PKBMetabolic abnormalities caused by a high sugar and high fat diet are becoming increasingly prevalent, and effective treatments are needed to prevent and manage these conditions.
