In vivo: To investigate the physiological effects of in-vivo inhibition of fatty acid synthesis on global lipid metabolism, a previous study administered cerulenin or C75 to mice. Profound weight loss following treatment was observed. Weight loss occurred dose-dependently and persisted for a duration. Moreoer, all treated mice recovered their lost body weight after the effect of the drug dissipated. The treatments of both cerulenin and C75 were well tolerated without obvious toxicity [2]. Cerulenin is a fatty acid synthase (FAS) inhibitor. Fatty acids are key aliphatic acids for energy production and storage, which are synthesized by a series of decarboxylative Claisen condensation reactions from acetyl-CoA and malonyl-CoA. In vitro: In a previous study, it was found that the inhibition of fatty acid synthesis at the physiologically regulated step of carboxylation of acetyl-CoA to malonyl-CoA was not cytotoxic to breast cancer cells. FAS inhibitor of cerulenin ould induce a rapid increase in intracellular malonyl-CoA to several fold, whereas 5-(tetradecyloxy)-2-furoic acid reduced intracellular malonyl-CoA by 60%.
