软肝宁对大鼠肝纤维化的保护作用研究木 闫冰,周晓辉,王曙东 (中国人民解放军南京军区南京总医院,江苏南京 210002) 摘要:目的探讨软肝宁对肝纤维化大鼠的影响,为临床适应证提供实验依据。方法将60只大鼠随机分为6组。正常对照组给予生理 盐水;模型组给予20%四氯化碳花生油溶液按10 mL/kg皮下注射,首剂加倍,每5 d注射1次,首次注射后,用生理盐水按10 mL/kg 灌胃,1次/天,连续6周;秋水仙碱组配成O.01 g/L;软肝宁高、中、低剂量组(0.12,0.06,0.03 g/kg)按损伤模型组同法给予CC1 造 模,然后给予软肝宁给药,1次/天。给药6周后检测大鼠血清中的丙氨酸氨基转移酶(ALT)、天门冬酸氨基转移酶(AST)、超氧化物歧 化酶(soo)、丙二醛(MDA)、血清透明质酸(HA)、层粘连蛋白(LN)、/1I型前胶原(PcⅢ)和羟辅氨酸(HYP),并观察大鼠肝组织常规病理 改变。结果软肝宁能改善肝纤维化大鼠的肝功能,高、中剂量可显著降低CCI 致大鼠肝损伤血清ALT,AST,MDA,HA,LN,PCIU水平和 肝组织中过高的HYP(P<0.05或P<0.O1),并能升高血清中SOD含量。常规病理显示,中、高剂量软肝宁可明显减轻CC1 所致的大 鼠肝细胞变性、坏死及肝组织损害程度。结论软肝宁具有延缓CCI 所致肝纤维化进程的作用。 关键词:软肝宁;肝纤维化;四氯化碳;大鼠 中图分类号:R285.5;R286 文献标识码:A 文章编号:1006—4931(2013)04—0012—03 Study on Protective Effects of Ruanganning Oil Rat Liver Fibrosis Yan Bing,Zhou Xiaohui,Wang Shudong (Nanjing General Hospital 0,’Nanjing Military Region,Nanjing,Jiaagsu,China 210002) Abstract:Objective To research the influence of Ruanganning on rats liver fibrosis to provide the experimental basis for its clinical indications.Methods 60 rats were randomly divided into 6 groups.The normal group was given with normal saline:the model group with 20%carbon tetrachloride peanut oil 1 0 mL/kg sc,the initial dose was doubled,once injection per 5 d,after initial dose,using norlTlal saline 10 mL/g ig,once daily for successive 6 weeks;the eolchicine group with colchicine 0.01 g/L;the high,middle and low Ruanganning groups(0.12,0.06,0.03 g/kg)were given CC14 for constructing the model by the method same to the lesion model group and then given Ruanganning,once daily.After 6一week medication,serum ALT,AST,SOD,MDA,LN,PC llI,HA,HYP were detected and the routine pathological changes of rat liver tissues were observed.Results Ruanganning could improve the hepatic function of rat live ̄’ fibrosis,especially the high and middle doses could significantly decrease the serum levels of AIJT,AST,MDA,HA,LN,PC 11I in CCI4~ induced liver damage rat and the exorbitant HYP in liver tissues(P<0.05 or P<0.01).Furthermore.the routine pathological ex— amination indicated that the middle and high doses of Ruanganning could alleviate CC14一induced liver cell degeneration,necrosis and the severity of fiver tissue damage.Conclusion Ruanganning is capable of delaying the progress of CCh—induced hepatofibrosis. 肝纤维化是一切慢性肝病的病理学基础,是形成肝硬化的必 经阶段”1。因此,抗肝纤维化已成为慢性肝病的重要治疗环节 ]。 软肝宁选用党参、姜黄、枸杞子、郁金、山芋肉等9味中药组方,长 期应用于临床。为探讨其作用机制,本研究中通过四氯化碳致肝 纤维化大鼠的影响”1,探讨软肝宁对肝纤维化大鼠的作用机制。 1 材料与方法 1 1 材料 药物及试剂:四氯化碳(CCh,分析纯,上海凌峰化学试剂有限公 司,批号为1 10524);秋水仙碱(昆明制药集团股份有限公司,批号为 201 10318);花生油采用普通食用花生油,试验时配成20%四氯化碳 花生油;软肝宁(选用党参、姜黄、枸杞子、郁金、山芋肉等9味中药组 成)按人临床剂量用体表面积法换算成大鼠用量后,分别研磨配制成 实验浓度,按每10g体重0.1 mL灌胃,1次/天,用时摇匀。AST试剂 盒(批号为20110312)、A 试剂盒(批号为20110426)、HA试剂盒 (批号为20110409)、LN试剂盒(批号为20110517)、PCⅢ试剂盒(批 号为201 10322)、HYP试剂盒(批号为201 1061 8j及ALB试剂盒(批 号为201 10322),均购自南京建成生物工程研究所。 动物:清洁级Wister雄性大鼠60只,体重210~230 g,由南 ・南京军区南京总医院课题,项目编号:2010M006。 京江宁县汤山青龙山动物繁殖场提供,实验动物生产许可证为 SCXK(苏)2010—2011。 1.2 方法 动物分组和造模方法:将60只大鼠随机分为6组,每组10只。 正常对照组(A组)给予生理盐水。模型组(B组)给予20%四氯化碳 花生油溶液按每10 g体重0.1 mL皮下注射,首剂加倍,每5天注射 1次,首次注射后,用生理盐水按每10 g体重0.1 mL灌胃,1次/天, 连续6周。软肝宁高、中、低剂量组(C ,C ,C,组)(0.12,0.06, 0.03 g/kg)按损伤模型组同法给予CCI 造模,然后给予软肝宁给 药,1次/天,连续给药6周。秋水仙碱组(D组)配成0.01 g/L按 60 kg体重10mg给药,1次/天,连续给药6周。 标本取材及预处理:各组动物在造模或给药后,以10%水 合氯醛麻醉,打开腹腔,颈总动脉取血6 mL,3 000 r/min 4 cIC 离心15 min后,分离血清,一2O℃保存待检。取肝脏,切取左侧 肝脏组织约5 mm x 5 mul x 3 mm大小投入10%甲醛固定用作 光镜观察。 1.3 观察指标 肝纤维化指标:用放射免疫法分别测定血清透明质酸(HA)、 ・药物研究・ Drug/dent/f/co#on

不同处方注射用甘草酸二铵的稳定性考察 陈爽,谢升谷,郑国钢 (浙江省食品药品检验所,浙江杭州 310004) 摘要:目的考察不同处方的注射用甘草酸二铵的稳定性。方法按2010年版《中国药典(二部)》附录《原料药与药物制剂稳定性试验指 导原则》进行影响因素试验,重点考察性状、溶液的颜色、pH、有关物质等评价指标。结果稳定性试验数据表明,不同处方的样品稳定性 存在较大差异。样品中有关物质随放置时间的变化因处方不同而呈现不同趋势。结论不同处方影响注射用甘草酸二铵的稳定性。 关键词:注射用甘草酸二铵;稳定性;影响因素试验 中图分类号:R927.11;R975 .5 文献标识码:A 文章编号:1006—4931(2013)04—0014—02 Investigation on Stability of Diammonium Glycyrrhizate for Injection with Diferent Formulas Chen Shuang,Xie Shenggu,Zheng Guogang (Zhejiang Provincial Institute dor Food and Drug Control,Hangzhou,Zhefiang,China 310004) Abstract:0bjective To investigate the stability of diammoniurn glycyrrhizate for injection with different formulas.Methods According to the stability testing of pharmaceutical preparations and guiding principles in the appendix of the Chinese Pharmacopoeia(edition 2010,Vo1.II),the influencing factors test was performed with the major investigation on the evaluation indexes of character,color of the solution,pH and related substances.Results The data of stability test indicated that the bigger difference existed in the stability of diammoniurn glycyrrhizate for injection with different formulas.The related substances in the samples of diammoniurn glycyrrhizate for injection with different formulas showed different trends with the change of storage time due to different formulas.Conclusion Different formulas affect the stability of diammoniurn glycyrrhizate for injection.
